چکیده

Among the microbial enzymes protease and amylase are most valuable which have been has diversified applications used extensively because of their capabilities in degradation organic wastes, application biofuels, agricultural, pharmaceuticals, chemical biotechnological industries. The aim current research work was purification, characterization alkaline proteases extracted from Bacillus cereus AUST-7. Various concentrations ammonium sulphate were applied for enzyme precipitation. Sephadex-G 100 FPLC system separation other proteins. SDS-PAGE to measure molecular weight required protease. Relative activities determined against different pH, temperature, incubation period activity. Stability temperature various metal ions inhibiter also studied. Purified on goat skin explore dehairing efficacy. A 6.5 purification fold 1163.50 U/mg specific activity obtained at 70% saturation 35. 91 8902 observed after separation. 35 kDa size exhibited SDS-PAGE. purified stable pH 10, 55 °C min period. found be 8–11, thermo-stability 50 phenyl methyl sulphonyl fluoride (PMSF) di-isopropyl fluorophosphates (DFP) inhibited good potential as agent leather

Isolation, Purification and Characterization of a Thermophilic Alkaline Protease from Bacillus subtilis BP-36

The goal of this research was to isolate and identify the thermostable alkaline protease producing bacteria among several native Iranian microorganisms. At the end of screening program, a Bacillus subtilis BP-36 strain producing thermophilic alkaline protease was isolated from a hot spring in Ardebil province. The target enzyme was purified using a one-step Aqueous two-phase systems (ATPS) prot...

Partial Purification and Characterization of Alkaline Protease from Bacillus sp. Isolated from Soil

Bacillus isolates were analyzed for alkaline protease production on casein containing agar plates and identified by clear zones of casein hydrolysis around colonies. Two gram positive Bacillus isolates S5 and C3 which showed the best enzyme production were studied for purification by precipitation and dialysis. The intracellular alkaline protease enzyme was purified about 4 fold with a yield of...
